The Cheetahs host the first Welsh franchise to visit South Africa, the Ospreys, in Bloemfontein.
Rhule will look to regain his lost form and confidence, after being axed from the Bok team in the wake of his horror show in Albany a fortnight ago – where he missed nine tackles in South Africa's 0-57 loss to New Zealand.
Rosco Specman will be providing cover on the bench.
The Cheetahs will be without a number of players through injury – Elandre Huggett (knee), Neil Jordaan (ankle and knee), Steven Meiring (appendix), Gerhard Olivier (ankle), Ruan van Rensburg (knee), Fred Zeilinga (hip) and Niel Marais (shoulder).
